4、 Precautions for using suppository molds
1. The suppository mold must be operated on a flat tabletop or ground.
2. Tighten the screws before operation to avoid weight differences in suppositories.

4. The suppository mold should be stored in a dry, ventilated, and clean place.

5、 The function of suppositories
1. Systemic effects
This type of suppository generally requires rapid release of drugs, especially antipyretic and analgesic drugs, which should be released and absorbed quickly. Oil based matrices, especially those with surface activity, are commonly used. They not only melt quickly at body temperature, but also disperse well. Generally, a matrix with opposite solubility to the drug should be selected based on its properties, which is beneficial for drug release and increased absorption. If the drug is lipid soluble, a water-soluble matrix should be chosen; If the drug is water-soluble, a lipid soluble matrix should be chosen. This method has a fast dissolution rate, high blood drug concentration, and short bee reach time. In order to improve the uniformity of drugs in the matrix, appropriate solvents can be used to dissolve or grind the drugs into fine powder before mixing with the matrix.
2. Local effects
This type of suppository only works locally in the cavity, and absorption should be minimized as much as possible. Therefore, a matrix that melts or dissolves slowly and releases the drug should be chosen. Suppositories made of water-soluble matrix have limited dissolution rate and slow drug release due to the limited amount of liquid in the cavity, which is more conducive to local efficacy than fatty matrix. For example, glycerol gelatin suppositories are commonly used as the matrix for local insecticidal and antibacterial vaginal suppositories.
6、 The preparation method of suppositories
There are generally three methods: kneading, cold pressing, and hot melting, which can be selected according to the different properties of the substrate. Generally, any of the three methods can be used for fatty matrices, while water-soluble matrices often use the hot melt method.
1. Rubbing and kneading method: Take the medicine and place it in a milk bowl (if it is a dry product, grind it into a fine powder first; if it is a paste, soften it with a small amount of suitable liquid first). Add an equal amount of matrix and knead it evenly. Then slowly add the remaining matrix and grind it as needed to form a uniform plastic mass. If necessary, an appropriate amount of vegetable oil or lanolin can be added to increase plasticity. Then place it on the porcelain plate, rub it with your hands across paper, gently press and rotate it, turn it into a cylindrical shape, and divide it into several equal parts according to the required amount, rub and knead it into the appropriate shape.
This method is suitable for the temporary preparation of small amounts of fat matrix suppositories. Its advantages lie in the fact that the preparation of suppositories does not require heating or melting, the drug is evenly dispersed in the matrix, no special equipment is required, and can be prepared in any situation. But the appearance of the obtained products is often inconsistent and not aesthetically pleasing.
2. Cold pressing method: This method uses instruments to press into suppositories and is suitable for mass production of fatty matrix suppositories. This method is similar to the kneading method. Take the medicine and place it in a suitable container, add an equal amount of matrix and grind it evenly, then add the remaining matrix and grind it evenly to make a block. After cooling, it is made into a file or granular form, and then installed in the tube of the suppository making machine. It is pressed into a certain shape through a model. To ensure the required amount of extrusion, it is often necessary to add 10% to 20% more raw materials, and the applied pressure must also be consistent.
There are many types of suppository presses, including horizontal and vertical ones, and their basic principles are the same. At present, this method is rarely used in production.
3. Hot melt method: This method is widely used, and suppositories with fatty and water-soluble matrices can be prepared using this method. Firstly, clean and dry the model. If necessary, use refined cotton or gauze dipped in a small amount of lubricant and apply it inside the model. Invert it to allow excess lubricant to flow out. Heat the calculated matrix powder on a water bath to melt, taking care not to raise the temperature too high. Then add the drug in different ways according to its properties, mix evenly, pour it into the mold until it slightly overflows the mold mouth, let it cool (it can be cooled in the refrigerator), wait for complete solidification, cut off the overflow part with a knife, open the model and push out the suppository. If there is excess lubricant on the suppository, it can be sucked off with filter paper.
There are two types of lubricants used in the preparation of suppositories. Oil based suppositories are commonly prepared with an alcohol solution consisting of 1 part of soft soap, 1 part of glycerol, and 5 parts of 90% ethanol. For suppositories with water-soluble or hydrophilic matrices, oil lubricants such as liquid paraffin and vegetable oil are used.
7、 Operation skills of suppository mold
1. After the suppository is made, the suppository mold containing the suppository can be placed in the refrigerator for cooling.
2. After the suppository mold is operated, it can be disinfected and cleaned with alcohol.
3. After the mold operation is completed, a layer of food grade lubricant can be applied on top, which can make the surface of the suppository smoother and crack free during the next operation.
